Anotace, referát	In the present invention there is disclosed a method for obtaining a biologically pure integrant of Bacillus thuringiensis, which produces at least 1.5 multiple of the quantity of a crystal delta-endotoxin and exhibits at least 1.5 multiple pesticidal activity, measured by killing or retarding of a pest growth, as compared to a crystal delta-endotoxin produced by a corresponding parental strain wherein the crystal delta-endotoxin produced by the integrant Bacillus thuringiensis integrant has an activity directed towards the same pest as the crystal delta-endotoxin produced by the corresponding parental strain, wherein the preparation process comprises (a) introducing into a cell of a parental strain a DNA construct lacking a Bacillus thuringiensis origin of replication comprising (i) a DNA sequence encoding a delta-endotoxin, wherein said delta-endotoxin is the same delta-endotoxin as the parental strain delta-endotoxin; (ii) a DNA sequence which is homologous with a region of the genome of said cell or said delta-endotoxin; and (iii) a selectable marker; (b) isolating the Bacillus thuringiensis strain with integrated DNA sequences of step (a) into the genome of said parental strain by homologous recombination in the presence of a selecting agent to obtain an integrant; and (c) amplifying the integrated DNA sequences in the selected Bacillus thuringiensis strain by cultivation of the strain in the presence of growing amounts of a selective agent and isolating the integrant from the culture of step (c). eng
Anotace, referát	Způsob získávání biologicky čistého integrantu Bacillus thuringiensis, který produkuje alespoň 1,5násobek množství krystalického delta-endotoxinu a vykazuje alespoň 1,5násobnou účinnost, měřeno usmrcováním nebo zbrzděním růstu škůdce, než má krystalický delta-endotoxin z odpovídajícího rodičovského kmene, přičemž krystalický delta-endotoxin produkovaný uvedeným integrantem je tentýž jako krystalický delta-endotoxin nativně produkovaný uvedeným rodičovským kmenem, kde: a) do genomu uvedeného rodičovského kmene Bacillus thuringiensis se integruje sekvence DNA kódující delta-endotoxin Bacillus thuringiensis, sekvence DNA, která je homologní s oblastí genomu uvedeného rodičovského kmene a selektovatelný signální znak, b) kmen Bacillus thuringiensis s integrovanými sekvencemi DNA ze stupně a) se izoluje, c) integrované sekvence DNA ve vybraném kmeni Bacillus thuringiensis ze stupně b) se amplifikují kultivací kmene v přítomnosti vzrůstajících množství selekčního činidla a integrant z kultury ze stupně c) se izoluje. cze
